Data treatment

The MesoSPIM is part of a more comprehensive imaging environment at Paris Saclay.

The specificities of the benchtop MesoSPIM make it an original equipment that completes the diversified light sheet instruments available in Paris Saclay. With a double-sided excitation path and a horizontal detection allowing rotation of the sample, acquisitions are performed in chambers of different dimensions adapted to aqueous immersion medium and solvent clearing solutions. The Axially Light Sheet Microscopy (ALSM) mode combined with the sCMOS camera rolling shutter mode ensures an homogeneous illumination over the whole field of view.

3 laser lines (488nm, 561nm and 638nm) are available.

Image data sets can be recorded in different formats (tiff, h5,...), stitched, and visualized in 3D using your favorite software. We use mostly ImageJ and Imaris.
